you can measure on top of the connector that's plugged into the motherboard.

put the red probe into pin1 (black wire), and the black probe into the next pin (pin2, grey wire) , you should measure a steady 12V when the drive is operating.

for the 5V, put the black probe into pin3 (grey wire), and the red probe into pin 4(grey wire, this is the opposite of the black wire), and measure for the 5V
